First bispecific antibody in Clinical Stage

First BiXAb® antibody, OT-A201, based on a dual immune checkpoint is currently being evaluated in a Phase 1 clinical trial, highlighting its potential as a novel immunotherapy approach in the treatment of solid tumors and hematologic malignancies.

Before going to clinical development, this first-in-class BiXAb® antibody has demonstrated not only good pairing, low aggregation, high stability, good manufacturability but also excellent specificity and safety, and displayed important in-vivo anti-tumoral activities at different doses with the potential for a wider therapeutic window as compared to its parental antibodies in preclinical studies.

OT-A201 was out-licensed to Onward Therapeutics for the treatment of hematologic and solid tumors. It is a BiXAb® targeting two immune checkpoints and has multiple modes of action.

Biomunex has also generated a best-in-class EGFR x HER3 bispecific antibody that was developed particularly in collaboration with University of Montpellier, to create new drugs for the treatment of pancreatic cancer. BMX-003 shows excellent activity both in vitro and in vivo in difficult models (Kras mutated) of pancreatic and colorectal cancer (some of these data are published in Rabia et al 2024).

Biomunex has a substantial pipeline of BiXAb antibodies at various stages of preclinical or clinical development that have the potential to treat many diverse cancer indications, both in solid tumors and haematological malignancies.

MAIT Engagers, a disruptive immuno-oncology approach

BIOMUNEX together with the Curie Institute in Paris, has invented the concept of using bispecific antibodies to bridge MAIT cells to cancer cells to elicit MAIT-mediated cancer cytotoxicity: MAIT engagers, a first-in-class innovative modality for the treatment of cancer. Using the BiXAb® platform technologies, Biomunex has generated a series of MAIT engagers that bridge MAIT cells to different tumor antigens in order to treat different cancer indications.

Classical CD3-Based T-Cell Engagers

BiXAb®-MAIT Engagers

Activation of ALL T-cell subsets
Only MAIT cells are activated
Overt cytokine release syndrome
No overt cytokine release —potentially safer in the clinic
Activation of regulatory T-cells (Tregs)
No increase in regulatory T-cell activity
Increased immunosuppression in the TME
Increased efficacy through preserved activity
Subsequent dampening of CD8 cytotoxicity

Biomunex has a substantial pipeline of MAIT engagers at various stages of development that have the potential to treat many diverse cancer indications.

The Series of BMX-500 programs, using Biomunex’s BiXAb® antibody platform, focuses on developing this novel disruptive and potentially universal MAIT engager approach, to specifically target and engage only MAIT cells and induce them to kill cancer cells. The specificity of this approach avoids cytokine release syndrome and the activation of regulatory T-cells in the tumor microenvironment and overcomes these limitations of classical CD3-based T-cell engagers.

2024 — LANDMARK LICENSING AGREEMENT

Biomunex has entered into an exclusive global licensing agreement with Ipsen for the clinical development of its first BiXAb®-MAIT engager, BMX-502.

Biomunex is eligible to receive up to $610 million  including an upfront payment already received

Bispecific antibody-drug conjugates

The ADC field has been reinvigorated with the success of ENHERTU® and several ADCs based on Mabs have been approved or are in late-stage clinical trials. The next wave of ADCs is using Bispecific antibodies to improve tumor targeting and reducing toxicity. The superb drug-like properties of the BiXAb platform render it a very suitable format for the next generation of bispecific antibody-drug conjugates. As such Biomunex is developing several projects with bispecific BiXAb-ADCs targeting several pairs of tumor antigens.
